Real-World Transition to Ublituximab From Prior Anti-CD20 Treatment in the Phase 4 ENABLE Study

Background: Ublituximab is a glycoengineered anti-CD20 monoclonal antibody approved for relapsing multiple sclerosis (RMS). ENABLE is an ongoing Phase 4 observational study evaluating its real-world effectiveness, safety, and tolerability; we report outcomes for participants who switched to ublituximab from a prior anti-CD20 therapy.
Methods: Participants who received at least one dose of ublituximab and had any baseline assessment as of the December 1, 2025 data cutoff were included. Annualized relapse rate (ARR) was calculated as cumulative relapses divided by cumulative treatment time, and patient-reported outcomes (TSQM and MSIS-29) were analyzed using mixed-model repeated measures.
Conclusions: Patients switching from prior anti-CD20 therapy to ublituximab demonstrated significant disease control with favorable safety and tolerability and improved patient-reported outcomes, consistent with the overall ENABLE population and pivotal trials.
